Does this build require the 3612_Ant-Cache.vp?

No.
However, if you don't have it, prepare for a lot of waiting while your cache files are being generated.

Would I be right in guessing that the Ant-6 shaders should probably be included?

Not necessarily. Those were test runs for the next set of mediavps shaders, if you like how they look, keep using them. But if you use the standard mvp shaders, you'll be fine as well.
