According to Karzai if there is a war between Pakistan and the US, he would support Pakistan (http://www.nytimes.com/2011/10/24/world/asia/karzai-says-afghanistan-would-back-pakistan-in-a-conflict-with-us.html)
KABUL, Afghanistan — Days after he stood with Secretary of State Hillary Rodham Clinton and accused Pakistan of harboring his country’s enemies, President Hamid Karzai of Afghanistan said this weekend that his country would support Pakistan if it ever went to war with the United States.
So after spending untold billions to put this guy into power and keep him there, he would ditch the only friend he has in favor of a country that plays such a big part of the security problems in Afghanistan. Now that's gratitude.....

Pakistan has never actually liked us. They just liked the gratuitous amounts of money that we've been stupidly pouring into the country.

Politics.
America (or rather, the people in power) wont give a ****, he knows that, so he's playing off on them to get the Pakistanis on side rather than risk an unstable border.

Hahahaha. . . Pakistan and Afghanistan going to war with the US.
-
It's just political posturing to sound good in the tribal regions that already care little for Pakistan and Afghanistan as it is right now.
-
Karzai hasn't been a friend of the United States for at least a half-decade, get with the goddamn program.

A war between Pakistan and India has a much higher possibility than a war between Pakistan and the USA.
And i'm quite sure that many of the northern Afghanis won't mind to fight at the side of the USA in a hypothetical US-Pakistani War. ;)

if karzai was a legit problem he wouldn't be in charge much longer. he also has very little control over his own country. he is the mayor of kabul, but certainly doesnt have a monopoly on violence with in his country. if push comes to shove he also doesnt have much of a military do anything with either at present. country is kind of in tatters. civil "strife" for half a century or more does that. imho its not really worth worrying about. hes just posturing.
EDIT: keep in mind the US is pulling out of the region while pakistan will be there for a while, and there is already concern over Pakistan shelling allied and afghan troops
EDIT2: also look at it from the afghans point of view. we came bringing war and conflict, not that life under the taliban was roses, and a lot of innocent afghans civilians have died. we are also meddling in their government again, which most peoples in general dislike. we also havent removed karzai even though its pretty apparent that the last election was rigged by/for him.

I think the average Afghani has probably had enough of war.
